Delivery remains the primary barrier to expanding oligonucleotide and peptide therapeutics beyond well validated tissues. This session will focus specifically on cross modality delivery approaches, where learnings from peptides, conjugates, lipids, and nanoparticle systems are combined to improve cellular uptake and tissue specificity. Key challenges include rational modality selection at discovery stage, balancing efficacy with safety, and managing complexity introduced by hybrid delivery systems. Discussion will address translational decision making, in vivo performance assessment, and early manufacturability considerations, with emphasis on how delivery strategy choices influence program progression and clinical risk.
