Poster Presentation: CMC and Process Development topic :Advancing siRNA Manufacturing through Chemical Fragment Synthesis and Enzymatic Ligation

The growing demand for siRNA therapeutics is driving the need for more efficient and scalable manufacturing strategies. Enzymatic ligation has emerged as a promising approach for joining chemically synthesized oligonucleotide fragments, while the quality and scalability of those fragments remain critical to overall process performance. This poster presents an integrated manufacturing concept in which solid-phase synthesis and liquid-phase synthesis serve as complementary options for fragment production prior to enzymatic ligation. We discuss how selecting the appropriate chemical synthesis platform can improve manufacturing efficiency and support future commercial production of increasingly complex siRNA therapeutics.
