Many high-value membrane protein targets, including GPCRs and ion channels, have remained untapped for therapeutic development due to technical barriers such as sequence conservation and structural complexity. We will discuss how advances in immunization and parallel engineering to optimize affinity, specificity, and developability are reshaping what is considered druggable. Drawing on case studies from campaigns against more than 100 previously inaccessible targets, we will also share key insights into target validation and preclinical candidate selection.
